Date palm micropropagation: Advances and applications Ciência e Agrotecnologia
Al-Khayri,Jameel Mohammed; Naik,Poornananda Madhava.
ABSTRACT Date palm (Phoenix dactylifera L.) is a fruit tree resilient to adverse climatic conditions predominating in hot arid regions of the Middle East and North Africa. The date fruit contains numerous chemical components that possess high nutritional and medicinal values. Traditional propagation by offshoots is inefficient to satisfy current demands for date palm trees. Alternatively, micropropagation provides an efficient means for large-scale propagation of date palm cultivars. Both somatic embryogenesis and organogenesis, either directly or indirectly though the callus phase, have been demonstrated in date palm in vitro regeneration. Culture initiation commonly utilizes shoot-tip explants isolated from young offshoots. Recently, the immature...

Development of blast resistant somaclones of the upland rice cultivar araguaia PAB
ARAÚJO,LEILA GARCÊS DE; PRABHU,ANNE SITARAMA; FREIRE,ADELSON DE BARROS.
The degree of blast resistance of upland rice (Oryza sativa L.) cultivar Araguaia has decreased over time causing significant yield losses. The major objective of this study was to obtain blast (Pyricularia grisea) resistant somaclones, adapting greenhouse and field selection procedures. Rice blast resistance and agronomic traits were assessed in R2 to R6 generations derived from regenerant plants (R1) from immature panicles of Araguaia. The evaluation and selection procedures include testing of early segregating populations and fixed lines in the advanced generations, under natural field conditions, and artificial inoculations in the greenhouse, with prevalent races IB-1 and IB-9 of P. grisea. Somaclones with both vertical resistance and slow blasting...

Genetic changes in grapevine genomes after stress induced by in vitro cultivation, thermotherapy and virus infection, as revealed by AFLP Genet. Mol. Biol.
Baránek,Miroslav; Raddová,Jana; Krizan,Bretislav; Pidra,Miroslav.
The Amplification Fragment Length Polymorphism (AFLP) technique was employed to study genetic variations which can be induced in vines by the stress occurring during different aspects of viticulture (in vitro cultivation, in vitro thermotherapy and virus infection). Analysis of AFLP banding patterns, generated by using 15 primer combinations, pointed to negligible genetic variation among plants exposed to individual stress. The average of similarity coefficients between differently stressed plants of the cultivars Müller Thurgau and Riesling were 0.984 and 0.991, respectively, as revealed by AFLP analysis. The low incidence of observed polymorphism demonstrates the high level of genome uniformity in plants reproduced by in vitro micropropagation via nodes,...

Genetic instability of sugarcane plants derived from meristem cultures Genet. Mol. Biol.
Zucchi,Maria Imaculada; Arizono,Hideto; Morais,Vicente Alberto; Fungaro,Maria Helena Pelegrinelli; Vieira,Maria Lúcia Carneiro.
The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subcultivations. Although different rates of polymorphism were observed, there was no direct association with the stage of subcultivation. The analysis of plants of two sugarcane varieties cultivated in vitro from meristems showed that variety RB83-5486...

Identification of rapd marker linked to blast resistance gene in a somaclone of rice cultivar Araguaia Trop. Plant Pathol.
ARAÚJO,LEILA G.; PRABHU,ANNE S.; FILIPPI,MARTA C..
The gene Pi-ar confers resistance to Pyricularia grisea race IB-45 in a somaclone derived from immature panicles of the susceptible rice (Oryza sativa) cultivar Araguaia. RAPD technique was used to identify molecular markers linked to this gene utilizing bulked segregant analysis. Initially, the two parental DNAs from the resistant donor SC09 and 'Araguaia' were analyzed using random primers. Of the 240 primers tested, 203 produced amplification products. The two parental DNAs along with the resistant and susceptible bulks of F2 population were screened using 48 primers that differentiated resistant and susceptible parents. Even though eight primers differentiated the resistant bulk from the susceptible bulk, as well as somaclone SC09 and 'Araguaia', only...

Indole-3-acetic acid metabolism in normal and dwarf micropropagated banana plants (Musa spp. AAA) Braz. J. Plant Physiol.
Zaffari,Gilmar Roberto; Peres,Lázaro Eustáquio Pereira; Tcacenco,Fernando Adami; Kerbauy,Gilberto Barbante.
Nanism is one of the most frequent type of mutant in micropropagated banana plants from the Cavendish subgroup. The present study aimed at studying some of the hormone factors involved in this type of mutation. Rhizomes from normal and dwarf plants from the cultivar Grand Naine were incubated for 5 d in the presence of [³H]-L-tryptophan, [³H]-indole-3-acetic acid and gibberellin, to quantify the endogenous levels of indole-3-acetic acid-ester, indole-3-acetic acid-amide, free indole-3-acetic acid, and cytokinins. The endogenous levels of indole-3-acetic acid and its ester- and amide-conjugated forms were measured in normal and dwarf plants incubated for 30 d with gibberellin, indole-3-acetic acid, and L- and D-tryptophan. In normal plants, the use of...

Molecular analysis of genetic stability in long-term micropropagated shoots of banana using RAPD and ISSR markers Electron. J. Biotechnol.
Lakshmanan,Venkatachalam; Reddampalli Venkataramareddy,Sreedhar; Neelwarne,Bhagyalakshmi.
A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Of the 4000 in vitro plantlets, 11 were used for screening that involved shoot cultures with distinct variation in morphological characteristics (morphotypes). Similarly, the mother maintained in the field was also subjected for genetic analysis. Out of the 50 RAPD and 25 ISSR primers screened, 30 RAPD and 5 ISSR primers produced totally 424 clear, distinct and reproducible band classes resulting in a total of 5088 bands where the banding patterns...

Somaclonal variation: a morphogenetic and biochemical analysis of Mandevilla velutina cultured cells BJMBR
Maraschin,M.; Sugui,J.A.; Wood,K.V.; Bonham,C.; Buchi,D.F.; Cantao,M.P.; Carobrez,S.G.; Araujo,P.S.; Peixoto,M.L.; Verpoorte,R.; Fontana,J.D..
Cell cultures of Mandevilla velutina have proved to be an interesting production system for biomass and secondary metabolites able to inhibit the hypotensive activity of bradykinin, a nonapeptide generated in plasma during tissue trauma. The crude ethyl acetate extract of cultured cells contains about 31- to 79-fold more potent anti-bradykinin compounds (e.g., velutinol A) than that obtained with equivalent extracts of tubers. Somaclonal variation may be an explanation for the wide range of inhibitor activity found in the cell cultures. The heterogeneity concerning morphology, differentiation, carbon dissimilation, and velutinol A production in M. velutina cell cultures is reported. Cell cultures showed an asynchronous growth and cells in distinct...

Somaclonal variation in Cereus peruvianus Mill. (Cactaceae): its potential to generate new varieties and broaden the species's genetic basis JBAG
Resende Gazoli,Adriana; Mangolin,Claudete Aparecida; Machado,Maria de Fátima.
In current study Random Amplified DNA-Polymorphism markers were used to detect genetic variations in C. peruvianus somaclones and to examine relationships among somaclones and plants grown from seeds in cultivated populations. The possible association of DNA fragments with morphological traits of somaclones was also investigated. Polymorphism in somaclones (62.24%) was higher than that detected in cultivated plants (42.57%). The casual grouping of somaclones presented atypical and typical shoot phenotypes showed that the random amplifi ed DNA-fragment polymorphisms detected in present study were not associated with shoot morphologies. Similarity in somaclones and cultivated plants ranged from 70.2 to 93.7% and 76.7 to 86.8%, respectively. Thus, in vitro...
